This is a working overview of placebo-controlled trial, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2026-06-09. Anything still debated is marked as such rather than presented as settled.
After subcutaneous administration, plasma concentrations rise within roughly thirty minutes and the elimination half-life is short, on the order of two to three hours. Reported physiological responses include transient increases in blood pressure and nausea, which tended to diminish with repeated dosing in trial settings. Because the peptide clears quickly, effects are not expected to persist long after a dose. Absorption from non-injected routes is poorly characterised, and nasal delivery produced variable plasma levels in older work.
Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.
Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
Receptor activation in hypothalamic and limbic circuits is the mechanism most often cited for the observed effects on sexual desire. Signalling through MC4R couples to Gs proteins and raises intracellular cyclic AMP, which in turn modulates dopaminergic tone in reward-related pathways. Because the peptide reaches the central nervous system after subcutaneous administration, peripheral vascular changes are regarded as secondary rather than primary. The precise neural circuits that translate receptor occupancy into behavioural change remain incompletely mapped, and published accounts describe the pathway in general terms rather than as a fully resolved sequence.

The lactam ring that closes the peptide backbone improves resistance to exopeptidase attack, but the molecule stays susceptible to hydrolysis and oxidation once dissolved. Degradation accelerates with temperature, extreme pH, and light exposure, and repeated freeze-thaw cycles promote aggregation and material loss. Lyophilized powder held desiccated at or below minus twenty degrees Celsius is the common way to keep reference material. Reconstituted solutions are generally kept cold and used within a short window because their stability is far lower than that of the dry solid.
Handling procedures emphasize minimizing moisture uptake, since the dried solid is hygroscopic and can draw water when warmed to room temperature before a vial is opened. Laboratories record lot number, purity, counter-ion content, and residual solvent data supplied by the producer. Impurity profiles are compared run to run, and any shift in retention time or peak shape prompts re-verification against a reference standard. Certificates of analysis are treated as claims to be checked rather than accepted at face value.
Across literature and commercial listings, the substance appears under several names, including bremelanotide, PT-141, and various brand designations. A CAS registry number provides a unique identifier for the compound, aiding retrieval from chemical databases. Synonym lists may also contain early development codes and descriptive chemical names. When consulting sources, researchers need to confirm that the cited names refer to the same molecular structure to avoid confusion. Naming consistency matters especially when comparing analytical data across publications.
PT-141 is a synthetic cyclic heptapeptide whose development code is bremelanotide. It belongs to the class of melanocortin receptor agonists and acts by mimicking endogenous peptide hormones. The compound originated from research on melanotan II, where investigators exploring derivatives found distinct pharmacological features. Unlike the parent compound, PT-141 showed effects on pathways related to sexual desire and function in early studies, prompting its development as a separate candidate. Its molecular design aimed to separate receptor activity from pigmentary effects.
Bremelanotide acts as a non-selective agonist at melanocortin receptors, with reported activity at MC1R, MC3R, MC4R and MC5R. The proposed basis for its central effects is activation of MC4R populations in the hypothalamus, a region associated with appetite and reproductive signalling. Because the peptide carries a net positive charge and polar side chains, it does not cross biological membranes freely, which is one reason oral administration is not the standard route. Effects generally appear within an hour of parenteral administration and are described as centrally mediated rather than peripheral.
Bremelanotide, developed under the code PT-141, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone. Its structure is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, with a lactam bridge joining the aspartate and lysine side chains. The molecule has the formula C50H68N14O10 and a monoisotopic mass near 1025 daltons. It is commonly prepared as the acetate salt and appears as a white to off-white lyophilised powder in solid form. The free acid is the pharmacologically relevant species, while the counter-ion improves handling and dissolution.
In September 2023, it was announced UPS had acquired the Long Beach, California-headquartered time-critical, health care logistics company, MNX for an undisclosed amount. In October 2023, it was announced UPS had acquired the Los Angeles-headquartered reverse logistics company, Happy Returns from PayPal for an undisclosed amount. In January 2024, UPS announced that it planned to cut 12,000 jobs and mandate that staff return to the office five days a week. Chief Executive Carol Tomé blamed the move on a "difficult and disappointing year" in 2023. In July 2024, UPS announced an agreement to acquire the Mexican logistics company Estafeta. Although the deal was expected to close in the first half of 2025, UPS terminated the planned acquisition on September 17, 2025, citing the inability to satisfy all required closing conditions in a filing with the SEC. According to reporting by The Wall Street Journal, the cancellation follows a more than year-long effort to integrate the Mexican small-package provider into UPS's regional growth strategy. In October 2025, the company announced it had reduced its workforce by 48,000 jobs in 2025, with 14,000 positions cut in management and 34,000 positions across its operations. In 2025, UPS began reducing the volume of packages it handled for its largest customer, Amazon, as part of a strategy to focus on higher-yielding business. The change was part of a broader restructuring of UPS's network and business portfolio.
Jerky is lean trimmed meat strips which are dehydrated to prevent spoilage and seasoned to varying degrees. Normally, this drying includes the addition of salt to prevent microbial growth through osmosis. The word "jerky" derives from the Quechua word ch'arki which means "dried, salted meat". Modern manufactured jerky is often marinated, prepared with a seasoned spice rub or liquid, or smoked with low heat (usually under 70 °C or 160 °F). Store-bought jerky commonly includes sweeteners such as brown sugar. Jerky is ready to eat, needs no additional preparation, and can be stored for months without refrigeration. A proper protein-to-moisture content is required in the final cured product to ensure maximum shelf-life. Many products that are sold as jerky consist of highly processed, chopped, and formed meat rather than traditional sliced whole-muscle meat. These products may contain more fat, but moisture content, as in the whole-muscle product, must meet a 0.75 to 1 moisture-to-protein ratio in the US. Jerky-like products can be found around the world, such as biltong in South Africa, pastirma in Turkey, kilishi in Nigeria and Cameroon, ch'arki (Quechua for dried, salted meat whose hispanicized spellings include charque, charqui or charquí) in South America, and cecina in Spain. The main processing districts of beef jerky in China are Inner Mongolia, Xinjiang and Yunnan.
If the metabolite only takes the right side new labeling patterns can occur, all in equal proportion. Other proportions can occur depending on how much of the original metabolite follows the left side of the pathway versus the right side of the pathway. Here the proportions are shown for a situation in which half of the metabolites take the left side and half the right, but other proportions can occur. These patterns of labeled atoms and unlabeled atoms in one compound represent isotopomers. By measuring the isotopomer distribution of the differently labeled metabolites, the flux through each reaction can be determined. MFA combines the data harvested from isotope labeling with the stoichiometry of each reaction, constraints, and an optimization procedure resolve a flux map. The irreversible reactions provide the thermodynamic constraints needed to find the fluxes. A matrix is constructed that contains the stoichiometry of the reactions. The intracellular fluxes are estimated by using an iterative method in which simulated fluxes are plugged into the stoichiometric model. The simulated fluxes are displayed in a flux map, which shows the rate of reactants being converted to products for each reaction. In most flux maps, the thicker the arrow, the larger the flux value of the reaction.

As a direct sampling technique, thermospray is able to gently ionize various types of analytes such that the resulting spectrum shows few fragments of the molecular ion and accompanying buffer gas components. This lack of fragmentation typically hinders the acquisition of structural information; however, thermospray is still capable of quantitative results and is valued for its range of viable analytes. When thermospray is coupled with high performance liquid chromatography mass spectrometry (TSP-HPLC-MS) the result is a highly sensitive method that is capable of lower detection limits than other HPLC-MS methods.

== Causes == DEB is caused by genetic defects (or mutations) within the human COL7A1 gene encoding the protein type VII collagen (collagen VII). DEB-causing mutations can be either dominant or recessive. Most families with family members with this condition have distinct mutations. Collagen VII is a very large molecule (300 kDa) that dimerizes to form a semicircular looping structure: the anchoring fibril. Anchoring fibrils are thought to form a structural link between the epidermal basement membrane and the fibrillar collagens in the upper dermis.
Mass spectrometry is also used to determine the isotopic composition of elements within a sample. Differences in mass among isotopes of an element are very small, and the less abundant isotopes of an element are typically very rare, so a very sensitive instrument is required. For the measurement of light elements (e.g. H,C,N,O,S), isotope ratio mass spectrometers usually use a single magnet to bend a beam of ionized particles towards a series of Faraday cups which convert particle impacts to electric current. A fast on-line analysis of deuterium content of water can be done using flowing afterglow mass spectrometry, FA-MS. Probably the most sensitive and accurate mass spectrometer for this purpose is the accelerator mass spectrometry. This is because it provides ultimate sensitivity, capable of measuring individual atoms and measuring nuclides with a dynamic range of ~1015 relative to the major stable isotope. Isotopic signatures can serve as markers of a variety of processes. Some isotope ratios are used to determine the age of materials for example as in carbon dating. Labeling with stable isotopes is also used for protein quantification (see protein characterization below).

More than a million earthquakes are recorded each year by a worldwide system of earthquake detection stations. The propagation velocity of the seismic waves depends on density and elasticity of the earth materials. In other words, the speeds of the seismic waves vary as they travel through different materials in the Earth. The two main components of a seismic event are body waves and surface waves. Both of these have different modes of wave propagation.

=== Tobacco === Polonium-210 in tobacco contributes to many of the cases of lung cancer worldwide. Most of this polonium is derived from lead-210 deposited on tobacco leaves from the atmosphere; the lead-210 is a product of radon-222 gas, much of which appears to originate from the decay of radium-226 from fertilizers applied to the tobacco soils. The presence of polonium in tobacco smoke has been known since the early 1960s. Some of the world's biggest tobacco firms researched ways to remove the substance—to no avail—over a 40-year period. The results were never published.
To develop organisms with novel or enhanced characteristics, this emerging field of study combines biology, engineering, and related disciplines' knowledge and techniques to design chemically synthesized DNA. Biomolecular engineering includes approaches that aim to create a toolkit of functional units that can be introduced to present new technological functions in living cells. Genetic engineering includes approaches to construct synthetic chromosomes or minimal organisms like Mycoplasma laboratorium. Biomolecular design refers to the general idea of de novo design and additive combination of biomolecular components. Each of these approaches shares a similar task: to develop a more synthetic entity at a higher level of complexity by inventively manipulating a simpler part at the preceding level. Optimizing these exogenous pathways in unnatural systems takes iterative fine-tuning of the individual biomolecular components to select the highest concentrations of the desired product. On the other hand, "re-writers" are synthetic biologists interested in testing the irreducibility of biological systems. Due to the complexity of natural biological systems, it would be simpler to rebuild the natural systems of interest from the ground up; to provide engineered surrogates that are easier to comprehend, control and manipulate. Re-writers draw inspiration from refactoring, a process sometimes used to improve computer software.

Trial results generally show a small to moderate average improvement in desire scores relative to placebo. The distribution of responses is wide, and some participants show little measurable change. Group averages should not be read as a prediction for any single person.
Nausea, flushing, headache, and transient blood pressure elevation are the most frequently reported events. These typically appear shortly after dosing and are usually described as mild to moderate in severity. Safety data covering long periods of continuous use remain limited.
A short half-life means the compound clears from circulation within hours, so any effect is tied closely to dosing time. Accumulation between doses is therefore unlikely under the studied schedule. The practical consequence is that timing of administration shapes what observers record.
The code designates bremelanotide, a cyclic heptapeptide melanocortin receptor agonist. It served as an internal research identifier before the compound entered formal clinical development. The code and the generic name refer to the same molecule.
