Bremelanotide is a synthetic cyclic heptapeptide developed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and appetite signalling. Its structure contains seven amino acid residues joined by a lactam bridge that closes the ring between two side chains. The molecular formula is C50H68N14O10 and the nominal molecular mass is near 1025 daltons. Much of the early laboratory literature refers to the same molecule by the development code PT-141.
The compound emerged from a research programme examining melanocortin analogues for effects on skin pigmentation. During early human studies, participants reported spontaneous erections as an unexpected side effect, which redirected development toward sexual function rather than tanning. An intranasal formulation was investigated in clinical trials but did not reach market approval. A subcutaneous injectable version later completed the regulatory process, and the nasal route does not appear in approved labelling.
Approved use is narrow and jurisdiction-specific. In the United States, the injectable product is authorised for premenopausal women with acquired, generalised hypoactive sexual desire disorder, a diagnosis that requires documented distress. It is not approved for men, for postmenopausal women, or for use alongside hormonal contraceptives under the approved labelling. Outside regulated markets, the same peptide is frequently sold as a research chemical, where identity, purity, and sterility are not independently verified.
Compared with the related compound melanotan II, PT-141 shows markedly weaker activation of receptors tied to pigmentation. This difference stems from subtle structural variations that alter affinity distribution across receptor subtypes. Investigators propose that such selectivity produces a different side effect profile in specific applications. However, downstream consequences of prolonged receptor activation remain uncertain in the literature. Published studies do not fully agree on the duration of signaling pathway activity and the mechanisms of desensitization.
From a pharmacokinetic standpoint, the peptide is usually delivered by injection because oral bioavailability is very low; proteases in the digestive tract degrade it rapidly. After subcutaneous administration, plasma concentrations reach a peak within roughly one hour. Its elimination half-life is relatively short, with most reports placing it in the range of a few hours. Nasal formulations have also been examined, though absorption varies widely between individuals. Metabolism proceeds mainly through peptidase cleavage, and the resulting products are excreted by the kidneys.
PT-141 initiates cellular signaling by binding to specific subtypes within the melanocortin receptor family. These receptors belong to the G protein-coupled receptor superfamily, and activation raises intracellular cyclic adenosine monophosphate levels. This cascade ultimately influences neuronal circuits in the central nervous system that are associated with sexual desire and arousal. Research indicates the compound's action concentrates in hypothalamic regions rather than peripheral tissues, which helps explain some observed pharmacological features. The selectivity of receptor binding underlies its functional differences.

How receptor activation translates into a change in desire is not established in detail. Proposed steps include modulation of dopaminergic signalling in reward circuits and downstream effects on autonomic tone. Human data consist mainly of clinical trials measuring self-reported outcomes rather than direct measurements of brain activity or transmitter release. The transient rise in blood pressure sometimes observed after administration is likewise reported consistently but explained only partly by known melanocortin pathways.
Melanocortin receptors form a family of five G-protein-coupled proteins, labelled MC1R through MC5R, that respond to peptides derived from pro-opiomelanocortin. Alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone are the best-known endogenous ligands. The receptors are distributed differently across tissues, so a single agonist can produce effects in the brain, skin, adrenal gland and vasculature. This distribution explains why one peptide can influence both pigmentation and motivated behaviour.
Bremelanotide is a cyclic heptapeptide that binds several melanocortin receptors rather than one. In vitro assays report agonist activity at MC1R, MC3R, MC4R and MC5R, with MC4R generally treated as the subtype most relevant to sexual effects. MC4R is expressed in hypothalamic nuclei involved in appetite, energy balance and motivated behaviour, which provides a plausible route for central action. Selective MC4R agonists studied in animals produce comparable behavioural changes, supporting that interpretation.
Polycythemia (also spelt polycythaemia) is a medical condition suggested by a laboratory finding that the hematocrit (the volume percentage of red blood cells in the blood) and/or hemoglobin concentration are increased in the blood, although this finding is also present in capillary leak syndrome. Polycythemia is sometimes called erythrocytosis, and there is significant overlap in the two findings, but the terms are not the same: polycythemia describes any increase in hematocrit and/or hemoglobin, while erythrocytosis describes an increase specifically in the number of red blood cells in the blood. Polycythemia has many causes. It can describe an increase in the number of red blood cells ("absolute polycythemia") or a decrease in the volume of plasma ("relative polycythemia"). Absolute polycythemia can be due to genetic mutations in the bone marrow ("primary polycythemia"), physiological adaptations to one's environment, medications, and/or other health conditions. Laboratory studies such as serum erythropoeitin levels and genetic testing might be helpful to clarify the cause of polycythemia if the physical exam and patient history do not reveal a likely cause. Mild polycythemia on its own is often asymptomatic. Treatment for polycythemia varies, and typically involves treating its underlying cause. Treatment of primary polycythemia (see polycythemia vera) could involve phlebotomy, antiplatelet therapy to reduce risk of blood clots, and additional cytoreductive therapy to reduce the number of red blood cells produced in the bone marrow.
== Episode 4: Looking for a Village == Kamiyah Morgan is a 6 year old little girl who suffers from a very unusual set of fainting episodes that will leave her unresponsive and immobile, and they can happen up to 300 times a day. When she experiences a fainting episode she will become completely paralyzed affecting everything in her body including her lungs, her mother states that every day that passes her ability to breathe diminishes. Kamiyah’s mother said that these fainting episodes started when she was about 8 months old as she would be crawling and suddenly tip over and go limp. At first their pediatric physician said the episodes looked like she was having a seizure, but after running an EEG there was no seizure being detected during the episodes. They then tried testing with MRI for any brain tumors or malignancies but again there was nothing. They were then referred over to the NIH or the National Institute of Health where their entire purpose is to be able to research and hopefully diagnose very strange cases. They NIH tested every system and every symptom and they were unable to find anything, they submitted her blood in for testing and they have yet to hear of anything from them for over two years. When Dr. Lisa Sanders asked the NIH for Kamiyah records; they were reluctant to release them even though she had her mother's permission, very shortly after this interaction Breteni, Kamiyah’s mother, received an email from NIH to discuss Kamiyah’s results.
Norway had rejected joining in a 1972 referendum, while the UK confirmed its membership in a 1975 referendum. Aside from the European Economic Community itself, the European continent underwent a profound transition towards democracy. The dictators of Greece and Portugal were deposed in 1974, and Spain's dictator died in 1975, enabling their accession in 1981 and 1986. In 1979, the European Parliament had its first direct elections, reflecting a growing consensus that the EEC should be less a union of member states, and more a union of peoples. The 1986 Single European Act increased the number of treaty issues in which qualified majority voting (rather than consensus) would be used to legislate, as a way to accelerate trade integration. The Schengen Agreement of 1985 (not initially signed by Italy, the UK, Ireland, Denmark or Greece) allowed movement of people without any border checks. Meanwhile, in 1987, the Soviet Union's Mikhail Gorbachev announced policies of "transparency" and "restructuring" (glasnost and perestroika). This revealed the depths of corruption and waste. In April 1989, the People's Republic of Poland legalised the Solidarity organisation, which captured 99% of available parliamentary seats in June elections. These elections, in which anti-communist candidates won a striking victory, inaugurated a series of peaceful anti-communist revolutions in Central and Eastern Europe that eventually culminated in the fall of communism.
In January 2023, Foreign Minister Mauro Vieira announced Brazil's return to the Community of Latin American and Caribbean States (CELAC). During the 7th CELAC summit, Lula expressed support for a modernization of the South American customs union Mercosur and the creation of a common unit of account between Argentina and Brazil which other South American nations could also join to boost regional integration and skip US dollar dominance. On 25 January, during a trip to Uruguay, Lula called for the European Union-Mercosur trade deal to be sealed and a China-Mercosur trade agreement to be explored. Lula and Uruguayan president Luis Lacalle Pou held talks over infrastructure projects to be developed in Uruguay, including a joint Uruguayan-Brazilian administration of the Rivera International Airport. In March 2023, Uruguayan and Brazilian top officials jointly detailed the said infrastructure projects in Brasília. Brazil also invited Paraguay and Uruguay to join the works of the G20 ahead of the 2024 G20 Brazil summit, as Brazilian rotating presidency in the group was to begin on 1 December 2023. In December 2024, during a summit in Montevideo attended by Lula and Ursula von der Leyen, Mercosur struck a definitive free trade agreement with the European Union representing nearly a quarter of the global gross domestic product (GDP).

An unknown Elamite king (sometimes speculated to be Khita) is recorded as having signed a peace treaty, in Old Elamite language written in an Old Akkadian ductus, with Naram-Sin (not deified in the text), stating: "The enemy of Naram-Sin is my enemy, the friend of Naram-Sin is my friend". Old Elamite is poorly understood (all other texts being very short) as yet making interpretation of the text challenging. The text mentions about twenty gods, mostly Elamite but with a few Sumerian and Akkadian, including Inshushinak, Humban, Nahiti, Simut, and Pinikir. It has been suggested that the formal treaty allowed Naram-Sin to have peace on his eastern borders, so that he could deal more effectively with the threat from Gutium.
Azidophenylalanine (4-azido-L-phenylalanine) is an unnatural amino acid derivative of L-phenylalanine, featuring an azide group at the para position of the phenyl ring. It is a bioorthogonal click-chemistry reagent that can be genetically incorporated into proteins via expanded genetic code techniques for site-specific labeling and functionalization. The compound serves as a vibrational reporter for local protein environments due to its azide group and is used in photo-crosslinking for protein interaction studies.
The human body's immune response to a surgically-implanted foreign object (prosthetic breast, cardiac pacemaker, orthopedic prosthesis) is to biologically isolate the foreign object with a capsule of tightly-woven collagen fibres. Afterwards, the capsular contracture occurs over time when the thickened, collagen-fibre capsule has compressed inwards, against the breast-implant with great mechanical pressure that deforms and breaks the implant, and so disfigures the breast; the causes of capsular contracture include bacterial contamination, shell-rupture of the medical device, leakage of the prosthetic filler-material, and hematoma. The prosthetic-breast implantation surgeries that have a low-rate of capsular contractures include surgical approaches that feature the submuscular emplacement of the breast-implant and the use of breast implants with a textured surface; limited handling of the breast implants before the surgery, limited contact with and handling of the skin of the implant-pocket, and irrigation of the surgical site with antibiotic solutions. To correct a capsular contraction, the plastic surgeon realises an open capsulotomy procedure to loosen and release the collagen-fibre capsule from the implant-pocket, for removal and replacement with a new model of prosthetic breast. Moreover, non-surgical therapies for treating collagen-fibre capsules include massage, external ultrasonic therapy, pharmaceutic therapy with leukotriene pathway inhibitor medications, and Diapulse therapy (Pulsed Electromagnetic Field Therapy, PEMFT).
=== West Asia === In Saudi Arabia, about 4.2% of the population carries the sickle cell trait, and 0.26% have sickle cell disease. The highest prevalence is in the Eastern province, where approximately 17% of the population carries the gene and 1.2% have sickle cell disease. In 2005, Saudi Arabia introduced a mandatory premarital test including HB electrophoresis, which aimed to decrease the incidence of sickle cell disease and thalassemia. In Bahrain, a study published in 1998 that covered about 56,000 people in hospitals in Bahrain found that 2% of newborns have sickle cell disease, 18% of the surveyed people have the sickle cell trait, and 24% were carriers of the gene mutation causing the disease. The country began screening of all pregnant women in 1992, and newborns started being tested if the mother was a carrier. In 2004, a law was passed requiring couples planning to marry to undergo free premarital counselling. These programmes were accompanied by public education campaigns.
=== Brain nonsynaptic glutamatergic signaling circuits === Extracellular glutamate in Drosophila brains has been found to regulate postsynaptic glutamate receptor clustering, via a process involving receptor desensitization. A gene expressed in glial cells actively transports glutamate into the extracellular space, while, in the nucleus accumbens-stimulating group II metabotropic glutamate receptors, this gene was found to reduce extracellular glutamate levels. This raises the possibility that this extracellular glutamate plays an "endocrine-like" role as part of a larger homeostatic system.

As an international coalition under United States command assembled in anticipation of an invasion of Iraqi-occupied Kuwait, the Iraqi regime decided to destroy as much of Kuwait's oil reserves and infrastructure as possible before withdrawing from the country. As early as December 1990, Iraqi forces placed explosive charges on Kuwaiti oil wells. The wells were systematically sabotaged beginning on January 16, 1991, when the allies commenced air strikes against Iraqi targets. On February 8, satellite images detected the first smoke from burning oil wells. The number of oil fires peaked between February 22 and 24, when the allied ground offensive began. According to the U.S. Environmental Protection Agency's report to Congress, "the retreating Iraqi army set fire to or damaged over 700 oil wells, storage tanks, refineries, and facilities in Kuwait." Estimates placed the number of oil well fires from 605 to 732. A further thirty-four wells had been destroyed by heavy coalition bombing in January. The Kuwait Petroleum Company's estimate as of September 1991 was that there had been 610 fires, out of a total of 749 facilities damaged or on fire along with an unspecified number of oil filled low-lying areas, such as "oil lakes" and "fire trenches". These fires constituted approximately 50% of the total number of oil well fires in the history of the petroleum industry, and temporarily damaged or destroyed approximately 85% of the wells in every major Kuwaiti oil field. Concerted efforts to bring the fires and other damage under control began in April 1991.

=== Centimorgan: Genetic linkage === In genetics, a centimorgan (abbreviated cM) or map unit (m.u.) is a unit for measuring genetic linkage. It is defined as the distance between chromosome positions (also termed loci or markers) for which the expected average number of intervening chromosomal crossovers in a single generation is 0.01. It is often used to infer distance along a chromosome. One centimorgan corresponds to about 1 million base pairs in humans on average.
In August 2008, Helton was diagnosed with a degenerative back condition, putting his health and ability to continue play in question. On May 19, 2009, Helton got his 2,000th hit, a single, during an 8–1 road loss to the Atlanta Braves. On July 22, Helton hit his 500th career double in a 4–3 home victory over the Arizona Diamondbacks. He became the 50th player in MLB history to hit 500 career doubles and the fastest to do since 1954. Helton also joined Babe Ruth, Stan Musial, Lou Gehrig and Ted Williams as the only players in MLB history to have at least 500 doubles, 320 home runs and a .325 batting average for a career. On March 11, 2010, Helton signed a two-year, $9.9 million contract extension, which ran through the 2013 season. Helton's degenerative back condition sent him back to the disabled list in July. He returned from the DL in August and hit .256 with 8 home runs and 37 RBI for the season. After the season, Helton said he would return to the Rockies in 2011, dispelling rumors of a possible retirement. On February 15, 2011, Helton announced his intention to play baseball for three more years, preferably for the Rockies. On June 30 he played his 2,000th career game. In 2012, Helton continued to deal with what was by now a chronic health issue. On April 14, he drove a 2-run walk-off home run through heavy rain, the seventh walk-off of his career. On July 13, Helton was placed on the 15-day disabled list due to inflammation in his right hip. He had played 63 games to that point, and was batting .235.

It is a synthetic cyclic peptide that acts as an agonist at melanocortin receptors. It is not a hormone replacement therapy and not a vasodilator in the usual clinical sense, although it does affect vascular tone. Chemically it belongs to the melanocortin analogue family.
PT-141 was the internal development code used during early research and it persists in older publications, conference abstracts, and informal listings. The code and the approved name refer to the same molecule. Its continued use reflects habit in the literature rather than any difference in substance.
No. The intranasal form was studied in trials but was never approved, and absorption through the nasal route varies considerably between individuals. Approved labelling covers a subcutaneous injection only. Any nasal product sold outside regulated channels falls outside verified manufacturing.
It primarily activates specific subtypes in the melanocortin receptor family. These receptors are G protein-coupled and mediate signaling mainly within the central nervous system.
