=== Pulp response to caries === Pulpal response to caries can be divided into two stages – pre- and post-infection. In caries-affected human teeth, odontoblast-like cells appear at the dentine-pulp interface along with specialized pulp immune cells to combat caries. Once they identify specific bacterial components, these cells activate innate and adaptive immunity. In uninfected pulp, leukocytes can sample and respond to the environment, involving macrophages, dendritic cells (DCs), T cells and B cells. This sampling process is part of the normal immune response, as it triggers leukocytes from the circulatory system to adhere to endothelial cells lining blood vessels and then migrates to the site of infection for defence. Macrophages can phagocytose bacteria and activate T cells, triggering the adaptive immune response that occurs in association with DCs. In the pulp, DCs secrete a range of cytokines that influence immune responses, and are key regulators of the infection defence. A comparatively small number of B cells are present in healthy pulp tissue, and pulpitis and caries progression increase their numbers. When bacteria get closer to the pulp but are still confined to primary or secondary dentine, acid demineralization of dentine occurs, producing tertiary dentine to help protect the pulp from further injury. After a pulp exposure, pulp cells are recruited and differentiate into odontoblast-like cells, contributing to the formation of a dentine bridge, increasing dentin thickness.

Emicerfont (GW-876,008) is a drug developed by GlaxoSmithKline which acts as a CRF-1 antagonist. Corticotropin releasing factor (CRF), also known as Corticotropin releasing hormone, is an endogenous peptide hormone which is released in response to various triggers such as chronic stress, and activates the two corticotropin-releasing hormone receptors: CRF1 and CRF2. This then triggers the release of corticotropin (ACTH), another hormone which is involved in the physiological response to stress. Emicerfont blocks the CRF1 receptor, and so reduces ACTH release. It has been investigated for the treatment of irritable bowel syndrome (IBS) and alcoholism, and while it was not effective enough to be adopted for medical use in these applications, it continues to be used for research, as the role of the CRH-ACTH system in IBS remains poorly understood.

Their reactions were a mix of hope and warning against manipulations, including views that the Cuban government could cease talks when it stabilizes itself. ==== Early synthesis by the Buddha ==== According to Erich Frauwallner, the twelvefold chain resulted from the Buddha's combination of two lists. Originally, the Buddha explained the appearance of dukkha from tanha, "thirst", craving. Later on, the Buddha incorporated avijja, "ignorance", as a cause of suffering into his system. This is described in the first part of dependent origination. Frauwallner saw this "purely mechanical mixing" as "enigmatical", "contradictory" and a "deficiency in systematization". Paul Williams discusses Frauwallner's idea that the 12 links may be a composite. However, he ultimately concludes that "it may be impossible at our present stage of scholarship to work out very satisfactorily what the original logic of the full twelvefold formula was intended to be, if there ever was one intention at all."
