It excels at matching remote homologs, particularly structures generated by ab initio structure prediction to structure families such as SCOP, because it emphasizes extracting a statistically reliable sub alignment and not in achieving the maximal sequence alignment or maximal 3D superposition. For every overlapping window of 7 consecutive residues it computes the set of displacement direction unit vectors between adjacent C-alpha residues. All-against-all local motifs are compared based on the URMS score. These values become the pair alignment score entries for dynamic programming which produces a seed pair-wise residue alignment.
== Pharmacokinetics and pharmacogenetics == It is bioactivated hepatically to its primary metabolite, N-desethylamodiaquine, by the cytochrome p450 enzyme CYP2C8. Among amodiaquine users, several rare but serious side effects have been reported and linked to variants in the CYP2C8 alleles. CYP2C8*1 is characterized as the wild-type allele, which shows an acceptable safety profile, while CYP2C8*2, *3 and *4 all show a range of "poor metabolizer" phenotypes. People who are poor metabolizers of amodiaquine display lower treatment efficacy against malaria, as well as increased toxicity. Several studies have been conducted to determine the prevalence of CYP2C8 alleles amongst malaria patients in East Africa, and have tentatively shown the variant alleles have significant prevalence in that population. About 3.6% of the population studied showed high risk for a poor reaction to or reduced treatment outcomes when treated with amodiaquine. This information is useful in developing programs of pharmacovigilance in East Africa, and have important clinical considerations for prescribing antimalarial medications in regions with high CYP2C8 variant frequency.
== Interactions == The CYP3A4 inhibitors ketoconazole and verapamil strongly increase exposure to gepirone, whereas lithium, paroxetine, and warfarin have no effect on exposure to gepirone. The CYP3A4 inducer rifampin profoundly decreases exposure to gepirone.

A quadcopter strike from Afghanistan on a cricket ground in Bajaur district, injured three civilians. On 2 May, Counter Terrorism Department claimed to have captured 21 suspected militants including those belonging to TTP in operations throughout Punjab. Pakistan claimed that 52 civilians had been killed in attacks from Afghanistan since the resumption of hostilities after the temporary ceasefire on Eid-ul-Fitr. On 3 May, a policeman was killed by militants in Lakki Marwat. A TTP commander was killed in an operation in Dera Ismail Khan. On 4 May, Pakistan reportedly launched an attack which resulted in the deaths of three civilians and the destruction of multiple schools, mosques, and a health center in Afghanistan. Pakistan disputed the claims. On the same day, a suicide attack foiled by security forces at a border checkpoint in Khyber Pakhtunkhwa resulted in a single casualty, seven civilians were also wounded. Afghan Taliban deployed hundreds of fighters and dozens of military vehicles to Spin Boldak, Shorabak and Registan districts along the Pakistan border in Kunar Province from other areas. On 6 May 2026, in Hangu district, security forces launched an operation after militants captured a police checkpost, a school and Shinwari Fort. On 7 May 2026, three children were killed and 10 civilians were wounded in a cylinder bombing by TTP and mortar attacks by Afghan Taliban in South Waziristan.

=== Pharmacodynamics === Sarcosine acts as a competitive inhibitor of GlyT1, a glycine transporter that is predominantly expressed on glial cells and is responsible for the reuptake of glycine from the synaptic cleft in the central nervous system. By blocking GlyT1, sarcosine elevates the extracellular concentration of glycine in the vicinity of NMDA receptors, thereby augmenting NMDA receptor-mediated neurotransmission. In addition to its indirect enhancement of NMDA receptor function via GlyT1 blockade, sarcosine directly acts as a co-agonist at the glycine binding site (also termed the GluN1 site) of the NMDA receptor. It increases NMDA-mediated currents in a dose-dependent manner. Sarcosine differs from glycine as a co-agonist in that it produces markedly less NMDA receptor desensitization at subsaturating concentrations. At equivalent receptor occupancy (EC20 to EC50), sarcosine significantly slowed the rate of glycine-dependent desensitisation compared with glycine itself, whereas the rate of glycine-independent desensitisation was similar for both ligands. Acetylcysteine or N-acetylcysteine (NAC) is a mucolytic that is used to treat paracetamol (acetaminophen) overdose and to loosen thick mucus in individuals with chronic bronchopulmonary disorders, such as pneumonia and bronchitis. It has been used to treat lactobezoar in infants. It can be taken intravenously, orally (swallowed by mouth), or by inhalation by use of a nebulizer. It is also sometimes used as a dietary supplement. Common side effects include nausea and vomiting when taken orally. The skin may occasionally become red and itchy with any route of administration. A non-immune type of anaphylaxis may also occur. It appears to be safe in pregnancy. For paracetamol overdose, it works by increasing the level of glutathione, an antioxidant that can neutralize the toxic breakdown products of paracetamol. When inhaled, it acts as a mucolytic by decreasing the thickness of mucus. Acetylcysteine was initially patented in 1960 and came into medical use in 1968. It is on the World Health Organization's List of Essential Medicines. It is available as a generic medication.
