All except cys 230 are essential for the catalytic process, forming multiple disulfide bridges during interaction with the substrates and regulators. == Treatment == The one known curative treatment is allogeneic stem cell transplantation, but this approach involves significant risks. Other treatment options are largely supportive, and do not alter the course of the disorder (with the possible exception of ruxolitinib, as discussed below). These options may include regular folic acid, allopurinol or blood transfusions. Dexamethasone, alpha-interferon and hydroxyurea (also known as hydroxycarbamide) may play a role. Lenalidomide and thalidomide may be used in its treatment, though peripheral neuropathy is a common troublesome side-effect. Splenectomy is sometimes considered as a treatment option for patients with myelofibrosis in whom massive splenomegaly is contributing to anaemia because of hypersplenism, particularly if they have a heavy requirement for blood transfusions. However, splenectomy in the presence of massive splenomegaly is a high-risk procedure, with a mortality risk as high as 3% in some studies. In November 2011, the US Food and Drug Administration (FDA) approved ruxolitinib (Jakafi) as a treatment for intermediate or high-risk myelofibrosis. Ruxolitinib serves as an inhibitor of JAK 1 and 2. Data from two phase III studies of ruxolitinib showed that the treatment significantly reduced spleen volume, improved symptoms of myelofibrosis, and was associated with much improved overall survival rates compared to placebo. However, the beneficial effect of ruxolitinib on survival has been recently questioned.

Collagen alpha-5(IV) chain is a protein that in humans is encoded by the COL4A5 gene. This gene encodes one of the six subunits of type IV collagen, the major structural component of basement membranes. Mutations in this gene are associated with X-linked Alport syndrome, also known as hereditary nephritis. Like the other members of the type IV collagen gene family, this gene is organized in a head-to-head conformation with another type IV collagen gene so that each gene pair shares a common promoter. Three transcript variants have been identified for this gene.

This culminated in the expulsion of the Jesuits from the Kingdom in 1769 and the forfeiture of their property. Even after Ferdinand came of age, Tanucci remained in office, but in 1774 he was exonerated at the instigation of Queen Maria Carolina of Austria, who wanted to bring Naples into the Austrian sphere of influence. However, the king continued season, pandering to the Neapolitan Enlightenment (Ferdinando Galiani, Antonio Genovesi and Gaetano Filangeri). During this period Calabria experienced major natural disasters, accompanied by social and economic changes: the plague arrived in 1743, striking Reggio Calabria and its surroundings from Messina, delaying the compilation of the cadastre onciario by local universities. The earthquake of 1783 struck southern Calabria killing some 50,000 and destroying Reggio, which was rebuilt according to more rational and linear architectural criteria.
